ABSTRACT

Objective: To investigate the gene frequency of HLA-B* 5801 in the population of Chinese Minnan region.Methods:In this study,we enrolled 178 patients requiring allopurinol therapy( including 40 patients with gout,89 patients with hyperuricemia and 49 patients with gouty arthritis) and 100 healthy people.We isolated genomic DNA from their blood and screened for HLA-B*5801 with both PCR and gene sequencing.Results:We found 22%patients and 16%healthy people with HLA-B*5801.The frequencies of HLA-B*5801 in patients and healthy people are 0.13 and 0.09,respectively.The results from PCR and gene sequencing were consistent.Conclusion:The frequency of HLA-B*5801 in the population of Chinese Minnan region is relatively high.Therefore,it is necessary to screen for HLA-B*5801 in allopurinol users before taking the medicine.

ABSTRACT

<p><b>UNLABELLED</b>This experiment was aimed to study the effects of different concentrations of medroxyprogesterone acetate (MPA) on proliferation, migration and apoptosis of endothelial progenitor cells (EPCs) in vitro and to provide experimental evidence for the use of MPA as anti-angiogenesis drug. We separated EPCs from bone marrow of a beagle dog and identified the expression of progesterone receptor (PR) in EPCs by immunocytochemistry. Proliferation tests (MTT analysis) were performed with EPCs in response to different concentration of MPA every 24 h for 7 consecutive days, the growth curve of these EPCs was obtained then. The inhibition rates were also obtained from MTT assay performed with EPCs exposed to different concentration of MPA. The migration tests were performed with EPCs in response to different concentrations of MPA. The cell cycle and apoptosis of EPCs exposed to different concentrations of MPA were analyzed by use of flow cytometry.</p><p><b>RESULTS</b>Immunohistochemical staining showed that EPCs were positive to PR. High concentration of MPA had significant inhibition effect on the growth of EPCs. This effect was time- and dose-dependent. But the low concentration of MPA had not have such effect, EPCs exposed to high concentration of MPA were found arrested in S phase, and the apoptosis rate increased. The migration ability of EPCs exposed to high concentration of MPA was also damaged.</p><p><b>CONCLUSION</b>High concentration of MPA can induce EPCs' apoptosis and inhibit their growth and migration. All these biological effects of MPA may be achieved through PR on EPCs.</p>

ABSTRACT

Objective To explore the curative effect of the pressure sore treated by iodophors associating with oxygen and peptide. Method 36 cases with pressure sore in stage Ⅲ and stage Ⅵ were randomly divided into the observation group and the control group. After ebriding was carried out in both groups, the control group was trea-ted with the rout change dressing and the other group was treated by iodophors associating with oxygen and peptide es-pecially. Result The cicatrization time of the observation group was remarkablely shorter than the control group. Conclusion Treating the pressure sore by iodopbors associating with oxygen and peptide has the advantage of less passion, shorter course of treatment, better curative effect, economy and mote convenience which is worth for the pro-moting of clinical usage.

ABSTRACT

Firstly, We used error-prone PCR to induce mutations on Armillariella tabescens MAN47 beta-mannanase gene, Secondly, we cloned the mutated fragments into secreted expression vector pYCalpha, Then the recombinant plasmids were transformed into Saccharomyces cerevisiae BJ5465 after amplified and extracted in DH5alpha cells. Through three cycles of error-prone PCR we built a mutant database, Then we screened one optimum (named M262) from about 104 mutants. The evoluted MAN47 beta-mannanase displayed both higher thermal stability and activity than wide type. The evoluted enzyme M262 retained high activity after treatment at 80 degrees C for 30 min, whereas, the wild type nearly lost activity under this condition. Meanwhile, the activity of M262 can reach to 25 U/mL, which is 4.3 times as wide type under optimum temperature. In addition, pH stability and pH range of evoluted enzyme M262 were both improved compared with wild-type enzyme. The optimum pH was estimated to be similar to that of wild-type enzyme. The sequence comparison illustrated that there were three nucleotide substitutions (T343A/C827T/T1139C) which carried corresponding amino acid changes (Ser115Thr/Thr276Met/Val380Ala). According to homologous modeling by SWISS-MODEL Repository, three mutated amino acids located at the sixth amino acid of the fourth beta-sheet, the first amino acid of the sixth alpha-helix, the turn between the tenth and eleventh beta-sheet, respectively.

ABSTRACT

<p><b>BACKGROUND</b>To evaluate the effects and toxicities of topotecan combined with cisplatin in the treatment of small cell lung cancer (SCLC).</p><p><b>METHODS</b>Twenty-six patients with SCLC diagnosed by pathologic or cytologic examination were treated with topotecan 1.25 mg/(m²*d) on days 1-5 and cisplatin 25 mg/(m²*d) on days 1-3. The chemotherapy was repeated every 21 days as a cycle.</p><p><b>RESULTS</b>Out of 26 evaluable patients, 3 achieved complete response and 11 achieved partial response with an overall response rate of 53.8%. The response rates were 75.0% and 35.7% in 12 previously untreated patients and 14 patients with prior chemotherapy, respectively. In addition, one of eight refractory patients and two of five patients with brain metastasis got partial responses. The median survival period was 27 weeks and one-year survival rate was 38.5%. Myelosuppression was the major dose-limiting toxicity.</p><p><b>CONCLUSIONS</b>Topotecan combined with cisplatin is effective for SCLC and it toxicity is tolerable.</p>
